No, it doesn't make any difference to their fitness if they play in an international or a Euro game, also they can't get injured so you don't have to worry about saving players for upcoming Euro games. Fitness levels are in effect in domestic cup games and they can get injured so you have to consider fitness levels ahead of these games and for upcoming league games, that's why I always say that you should build a squad of about 25 before you sign any expensive big name players.
