Is not match fit applied after a game? or does it occur during a game?

Is the tiredness applied after the game is over or is it applied to the players during the game? I have some players that are close to nmf. Just want to find out if the will become nmf during a game... and need to be subbed?
Answers
Such players will become tired (nmf) after the match, best thing to do is to not select them but rest them for the next match.
